The county formed on January 4, 1825, from sections of Coshocton, Tuscarawas, and Wayne counties, named after War of 1812 officer Andrew Holmes.
Nearly half of the county's 44,223 residents belong to the Amish community, maintaining traditional farming practices and Pennsylvania German language.

Current demographic trends indicate Holmes County will become the first county in the United States with an Amish majority by 2030.
