Pope Leo X authorized the hospital's establishment in 1514, with Martín de Jerez founding it in 1515 as a successor to Hospital de Nuestra Señora de la Antigua.
The main altarpiece displays La Piedad sculpture, depicting the Virgin Mary holding her deceased son after the crucifixion descent.
The religious services are conducted by the Congregation of Saint Vincent de Paul, which has managed the temple since 1923.
During the bubonic plague epidemic of 1582, the hospital reached maximum capacity, leading to the opening of additional medical facilities in the city.
