The Museu do Rio Cuiabá occupies the 1899 Mercado do Peixe building, a neoclassical structure of locally available materials in the Porto neighbourhood. Its galleries hold photographs, handicrafts, sacred art and scale models of the Cuiabá River region, and it is listed as state heritage.

The building dates from 1899 and was the Mercado do Peixe, also called Mercado do Porto, an open-air fish fair that on Sundays filled the riverside streets around an older, poorly kept structure, serving riverside dwellers and fishermen. The state government listed it in 1983, and from 1996 the municipality restored it as the Museu do Rio Cuiabá, moving the fair to a large shed.

The collection displays objects from daily life of river-dependent communities and religious art that mirrors how the city developed in relation to the waterway. Visitors can see how closely the rhythm of life in Cuiabá has always been tied to this river.
