The cathedral was built between 1935 and 1950 and was consecrated in 1936, making it Australia's first purpose-built Russian Orthodox church. It served as the seat of the Russian Orthodox Diocese from 1948 to 1950 under Bishop Theodore Rafalsky's leadership.

The cathedral serves as a spiritual center for the Russian Orthodox community in Brisbane, where traditional liturgical practices continue to shape the daily faith life of worshippers. Services in Church Slavonic and English reflect the place's role as a bridge between Russian heritage and Australian society.
